Don't use the blend tool for one. Try using the airbrush to put the highlights in instead.

i think we'd almost have to see what you've done before we could give any real useful feedback.

For my opinion , I think you should always go with the flow of the line art .
How i do it is simple .
1st - I generate a global shading of the area that i want to color . ( With the airbrush and smudge tool )
2nd - I sperate each muscle with the same tech as for the global shading .
3rd - I add the highlight but always keep in mind the light source effect on each part you are coloring ( meaning the intesity of the highlight must be carefuly ajusted ) tool : ( airbrush , smudge tool )
4th - I create some texture , depending on what of the feel i want to give to the area colored . tool : ( airbrush , smudge tool )
Hope it helped ya .
well i use cuts.
look up laura's tutorials about cuts here

im no experrt but when i color muscles i use the polygonal lasso too to isolate what i want colored tehn i basicaly shade it dark onaround the outside and add light near the center, then add hilites for the the light source, then if anything looks disconnected i just use the smudge tool
